Dr Reddy was reported missing on Sunday after she failed to return home following a dental convention at St Leonards over the weekend.

She was captured on CCTV inside the restaurant and appeared to leave alone a short time later.

At about 9.30pm on Tuesday, officers located a auto parked in Strachan Street, Kingsford.

Officers found the man's 32-year-old ex-girlfriend's body in her vehicle on Strachan Lane in Kingsford about 9.30pm on Tuesday night.

"Inquiries by officers from Sydney City Police Area Command are continuing into the circumstances surrounding the woman's death", a police statement said.

Her family didn't know if she attended the parade, where she stayed on Saturday night, where she was on Sunday morning, or what happened to her auto.

Investigators said they have since been notified that the man later died in a head-on collision on the New England Highway in Willow Tree, just outside of Tamworth, about 10pm on Monday night.

Police released the last images they had of Reddy at a McDonald's on George Street in the city centre after Mardi Gras on Sunday morning at 2.15am.

A Facebook page was set up soon after Dr Reddy went missing, calling for people to "help find Preethi Reddy".

One resident, who did not want to be named, said she returned home at 9.45pm last night and saw most of the street was blocked off by police.

The man is believed to be her former boyfriend who was also attending the dental conference, a police source told AAP.

Investigations are ongoing. Anyone with information on the matter has been urged to call Crime Stoppers on 1800 333 000.

Preethi's disappearance is "out of character" and there are serious concerns over her welfare, a New South Wales (NSW) Police spokeswoman said.

Reddy had been a dentist for ten years, according to a professional dentistry website, and is described as having a specific interest in children's dentistry.
